Objective To explore the expression and clinical significance of β-catenin and E-cadherin in colorectal cancer. Methods Immunohistochemistry was employed to detect β-catenin and E-cadherin expression in 60 cases of colorectal cancer and corresponding normal mucosa. The relationship between protein expression of β-catenin, E-cadherin and clinicopathologic characteristics in colorectal cancer was studied. Results In 60 cases of colorectal cancer, the abnormal expression rates of β-catenin and E-cadherin proteins were 75. 00% and 58. 33% respectively, significantly higher than in corresponding adjacent normal colorectal mucosa (1. 67% , 6. 67% ; P<0. 01). The abnormal expression of β-catenin and E-cadherin proteins showed a positive correlation (r = 0. 683, P<0. 01). The abnormal expression of β-catenin and E-cadherin proteins was correlated to tumor differentiation grade, lymph node metastasis and Dukes stage (P<0. 05). In addition, E-cadherin was also correlated to tumor histological type(P<0. 05). Conclusion The protein expression of β-catenin and E-cadherin is abnormal in colorectal cancer. E-cadherin can indirectly affect the Wnt signaling pathway conduction by regulating β-catenin. Joint detection β- catenin and E-cadherin proteins may provide certain molecular basis to for prevention early diagnosis and new target treatment of colorectal cancer.%目的 探讨β连环蛋白(β-catenin)和E-钙粘蛋白(E-cadherin)在结直肠癌组织中的表达及其临床意义.方法 应用免疫组织化学染色技术(Elivision)检测β-catenin和E-cadherin蛋白在这60例结直肠癌及癌旁正常黏膜中的表达情况及其与临床病理特征的关系.结果 60例结直肠癌组织中,β- catenin、E-cadherin 蛋白的异常表达率分别为75.00%、58.33%,显著高于癌旁正常结直肠黏膜组织1.67%、6.67%(P<0.01).β-catenin与E-cadherin蛋白的异常表达呈正相关(r= 0.683,P<0.01).β-catenin 和E-cadherin蛋白异常表达均与肿瘤的分化程度、淋巴结转移和Dukes分期有关(P<0.05).此外,E-cadherin蛋白表达还与肿瘤的组织学类型有关(P<0.05).结论 β-catenin和E-cadherin在结直肠癌中均有异常表达,E-cadherin可通过对β-catenin的调节,间接地影响Wnt信号通路传导.两者联合检测可能为结直肠癌的预防、早期诊断、新靶点治疗提供新思路和分子依据.
