Objective To establish optimal method for the infection of recombinant adenovirus (Ad) in vivo. Methods The different dose of Ad suspension was injected into the carotid artery for different times after blockade of blood stream in rats. The infection rate was tested by immunohistochemistry. Results Optimal infection was showed at 20 μl of Ad suspension for 20 min. Conclusions The infection rate is positive correlative with dose and time of Ad infection to certain extent.%目的 探讨重组腺病毒在体高效感染血管组织的优化条件.方法 将制备的KLF4腺病毒悬液,以不同剂量(10、20、40 μl)导入大鼠颈总动脉腔内,停留不同时间(10、20、30 min).感染后48 h用免疫组化染色评价感染效率.结果 采用20 μl-20 min的感染条件,既可实现较理想的感染效率,又不破坏血管组织的完整性.结论 一定条件下腺病毒的剂量和感染时间与感染效率呈正相关关系.
