芡实提取物对D-半乳糖衰老小鼠学习记忆障碍的改善作用

         

摘要

Objective To investigate the effect of different extracts from Euryale ferox seed on D-galactose inducing aging mice and its mechanism. Methods Kunming mice were randomly divided into control, model, low and high doses of ethanol, acetic ester and normal butanol extracts from Euryale ferox seed groups. Open field device and the Morris water maze device were used to test independent activities and the learning and memory abilities. Nitric oxide synthase (NOS) , glutathione peroxidase ( GSH-Px) , cholinesterase (AChE) activities in brain tissue were detected. Results The ethanol, ethyl acetate and normal butanol extracts from Euryale ferox seed could dose-depend-ently improve learning and memory, NOS and GSH-Px abilities of D-galactose inducing aging mice, and reduce AChE activity. Conclusions The ethanol, ethyl acetate and normal butanol extracts from Euryale ferox seed can play the function of delaying senility, which may be related to promoting the body clearing lipid peroxide, enhancing antioxidant capacity of brain tissue and improving the role of cholinergic system in learning and memory. The effect of normal butanol extract is better than that of ethanol extract and the effect of ethanol extract is better than that of ethyl acetate extract.%目的 观察芡实不同提取物对D-半乳糖致衰老小鼠学习记忆能力的影响及其作用机制.方法 将昆明种小鼠随机分为8组,即对照组、模型组、芡实乙醇提取物高低剂量组、芡实乙酸乙酯提取物高低剂量组、芡实正丁醇提取物高低剂量组.分别采用旷场装置和Morris水迷宫装置测试各组小鼠的自主活动状况及学习记忆能力,同时测定各组小鼠脑组织一氧化氮合成酶(NOS)、谷胱甘肽过氧化物酶(GSH-Px)、乙酰胆碱酯酶(AChE)的活性.结果 芡实乙醇提取物、乙酸乙酯提取物、正丁醇提取物均能浓度依赖性地改善亚急性衰老小鼠的学习记忆能力,提高脑组织NOS、GSH-Px活力,降低AChE活力.结论 芡实乙醇、乙酸乙酯、正丁醇提取物具有改善小鼠衰老状态的作用,其机制可能与其能促进机体清除脂类过氧化物,增强脑组织抗氧化能力,改善胆碱能系统在学习和记忆中的作用有关.芡实正丁醇提取物的作用优于乙醇提取物,乙醇提取物的作用优于乙酸乙酯提取物.
