三硝基苯磺酸/乙醇与结肠细菌菌株诱导的实验性结肠炎模型的建立与评价

         

摘要

Objective To establish the models of closer to people in the natural course of IBD through comparing trinitrobenzene sulphonic acid (TNBS)/ethanol and rat Colibacillus strain-induced colitis models, and further explore the IBD pathogenesis and lesion development law. Methods Fifty female Sprague-Dawley (SD) rats were randomly divided into three groups: normal control group ( group N, n = 20), TNBS/ethanol group ( group A, n = 20) , and rat Colibacillus strain-induced colitis group (group B, n = 20). The colitis models of rats were established with TNBS/ethanol and rat colonic bacterial strain. Survival rate and disease activity index (DAI) were recorded. The colonic macroscopic damage index (CMDI) and tissue damage index (TDI) were observed in the 14th and 28th days after model established. Resuits Two rats died in the group A. The DAI, CMDI and TDI were significantly higher ( all P < 0. 05 ) in model groups than those in normal group. The DAI, CMDI and TDI had no significant difference in the 14th and 28th days of model groups, and between the two model groups. Conclusion The model of rat Colibaeillus strain-induced colitis is closer to the natural course of IBD from the development process and pathological mechanism of the IBD, which is a good model to knowledge pathogenesis of IBD and evaluate the therapy effect of drugs.%目的 比较2,4,6-三硝基苯磺酸 (2,4,6-trinitrobenzene sulphonic acid,TNBS)/乙醇和大鼠结肠细菌菌株诱导两种结肠炎模型,建立更加接近人IBD自然病程的模型,进一步探讨IBD发病原因及病变发展规律.方法 50只SPF级雄性Sprague-Dawley (SD)大鼠随机分为正常对照组(N组,20只),TNBS/乙醇模型组(A组,20只)、大鼠结肠细菌菌株模型组(B组,20只).观察造模后存活率,每日行疾病活动指数评分(disease activity index,DAI),分别于造模后第14天、第28天各处死一半大鼠,评价结肠大体形态损伤指数(colon macroscopic damage index,CMDI)及组织学损伤指数(tissue damage index,TDI).结果 A组2只大鼠死亡,模型组DAI、CMDI及TDI评分明显高于正常对照组(均P<0.05),模型组于第14天与第28天处死大鼠DAI、CMDI及TDI评分及模型A、B组之间无显著性差异.结论 从病变发展过程、病程及病理改变情况上看采用大鼠的正常菌群为抗原,比较接近于自然情况,可作为研究IBD发病机制及评估药物疗效的一个良好模型.
