脐血干细胞经不同途径移植对大鼠脊髓损伤运动功能的影响

摘要

Objective To investigate the effect of umbilical cord blood stem cell transplantation via different approaches on motor function in spinal cord injured rats.Methods The models of spinal cord contusion injury in 80 SD rats were established by the modified Allen's weight dropping method.They were divided into four groups and randomly and equally.The rats in group A were treated with local umbilical cord blood stem cell transplantation at 7th day after injury.The rats in group B were treated with umbilical cord blood stem cell vein transplantation at 7th day after injury.The rats in group C were treated with umbilical cord blood stem cell lumbar puncture transplantation at 7th day after injury.The rats in group D were treated with equal volume phosphate buffer (PBS) solution at 7th day after injury.The number of cells in groups A,B and C was all 20 × 105.Basso-beattie-bresnahan (BBB) scale and inclined plane test were performed to assess the motor function changes of rats before and 7,14,21,28,35,42 days after operation.Results Scores of BBB scale (group A:2.05 ± 0.39,4.05 ± 0.51,7.40 ± 0.88,9.95 ± 0.83,12.00 ±0.97,14.15 ±0.75;group B:2.15 ±0.37,4.15 ±0.49,7.55 ±0.89,10.05 ±0.83,12.30 ±0.86,14.45 ±0.76;group C:2.10 ±0.48,4.05 ±0.60,7.45 ±0.76,10.10 ±0.64,12.15 ± 0.75,14.35 ± 0.67) and inclined plane test (group A:30.00 ± 2.81,44.75 ± 2.55,48.25 ± 3.35,50.50±3.20,58.25 ±4.38,68.50 ±3.28;group B:30.25 ± 1.97,44.25 ±2.94,48.75 ±3.58,50.25 ±3.43,59.50 ± 3.59,69.00 ± 3.84;group C:29.75 ± 2.55,43.75 ± 3.58,50.25 ± 3.80,50.75 ± 3.35,60.00 ± 3.63,68.00 ± 3.77) were higher in groups A,B and C than in group D (Scores of BBB scale:0.90±0.31,2.05 ±0.39,2.55 ±0.51,3.80 ±0.52,4.10 ±0.55,5.10 ±0.64;inclined plane test:25.25 ± 3.02,30.50 ± 2.24,31.50 ± 3.28,37.25 ± 4.99,42.50 ± 3.03,44.50 ± 2.24).Scores of BBB scale and inclined plane test in groups A,B and C were higher than those in group D,and the difference was statistically significant at the 28th,35th and 42nd day (P =0.000).Scores of BBB scale and inclined plane test were not significant better between groups A and B,groups A and C,and groups B and C.Conclusion Transplantation of umbilical cord blood stem cell may achieve behavioral improvement for the injured spinal cord.There was no significant difference among the three types of umbilical cord blood stem cell transplantation.%目的 观察脐血干细胞经不同途径移植对大鼠脊髓损伤的运动功能影响.方法 改良的重物打击法制作SD大鼠急性脊髓损伤动物模型80只,随机等分成4组.损伤7d后实施手术,A组为干细胞局部移植组,损伤后给予20×105个脐血干细胞局部移植;B组为干细胞静脉移植组,损伤后给予20×105个脐血干细胞静脉移植;C组为干细胞腰穿移植组,损伤后给予20×105个脐血干细胞腰穿移植;D组为对照组,损伤后给予同样体积的磷酸盐缓冲液(PBS)处理.分别于损伤前及损伤后的7、14、21、28、35、42 d,通过脊髓损伤运动功能(BBB)评分及斜板实验评估大鼠后肢运动功能的恢复情况.结果 A、B、C3组大鼠在脊髓损伤后第7、14、21、28、35、42天的BBB评分(A组:2.05 ±0.39、4.05 ±0.51、7.40 ±0.88、9.95 ±0.83、12.00±0.97、14.15±0.75;B组:2.15±0.37、4.15±0.49、7.55±0.89、10.05±0.83、12.30±0.86、14.45±0.76;C组:2.10±0.48、4.05±0.60、7.45±0.76、10.10±0.64、12.15±0.75、14.35±0.67)及斜板实验评分(A组:30.00±2.81、44.75±2.55、48.25±3.35、50.50±3.20、58.25±4.38、68.50±3.28;B组:30.25±1.97、44.25±2.94、48.75±3.58、50.25±3.43、59.50±3.59、69.00±3.84;C组:29.75±2.55、43.75±3.58、50.25±3.80、50.75±3.35、60.00 ±3.63、68.00±3.77)都高于D组(BBS评分:0.90±0.31、2.05±0.39、2.55 ±0.51、3.80±0.52、4.10±0.55、5.10±0.64;斜板实验评分:25.25±3.02、30.50±2.24、31.50±3.28、37.25±4.99、42.50±3.03、44.50±2.24).A、B、C3组大鼠在28、35、42 d的BBB评分及斜板实验评分高于D组的差异有统计学意义(P =0.000).A、B、C3组大鼠之间的BBB评分和斜板实验评分比较,差异均无统计学意义.结论 脐血干细胞移植可以促进脊髓损伤大鼠运动功能的恢复.3种脐血干细胞移植方式之间没有明显的临床差异.
