γ干扰素对甲状腺刺激抗体作用下FRTL5细胞TG、TPO基因表达的影响

摘要

目的 观察γ干扰素对甲状腺刺激抗体(TSAb)作用下FRTL5细胞甲状腺球蛋白(TG)、甲状腺过氧化物酶(TPO)基因表达的影响。方法 本研究运用Graves病患者血清经聚乙二醇4000处理获得TSAb的粗提物,用3H掺入法及Northern印迹杂交法观察γ干扰素对TSAb作用下FRTL5细胞生长及TG、TPO基因表达的影响。结果 TSAb上调FRTL5细胞的生长和TG,TPO基因的表达,而γ干扰素对TSAb诱导作用下的FRTL5细胞生长及基因转录(TG、TPO mRNA)均起抑制作用。结论 γ干扰素可抑制Graves病的自身抗体TSAb对甲状腺细胞生长和功能的刺激作用,推测γ干扰素在Graves病的发病中起一定的调节作用。%Objective To observe the effects of interferon γ (IFNγ) on thyroglobulin (TG) and thyroid peroxidase (TPO) gene expressions in FRTL5 cells induced by thyroid-stimulating antibody (TSAb). Methods TSAb crude fraction was extracted by polyethylene glycol 4000. Recombinant rat interferon-γ (0, 1, 10, 102, 103 U/ml) was added to the FRTL5 cells induced by TSAb, and then expressions of TG, TPO mRNA were measured by Northern blot, the cell growth was measured by 〔3H〕 thymidine incorporation. Results (1)TSAb increased TG, TPO gene expressions, and 〔3H〕 thymidine incorporation. (2)Interferon-γ  inhibited  TSAb-induced  TG, TPO  gene  expressions  and  〔3H〕 thymidine incorporation. Conclusion Interferon-γ inhibits the growth and function of thyrocytes induced by TSAb, which suggests that interferon-γ might regulate thyroid function in Graves' disease.
