骨形成蛋白在小白鼠体内诱导成骨的测定方法

         

摘要

Objective To search for a new method to determinate the boneremodeling ability of bone morphogenetic protein( BMP).Methods Self-prepared bovine morp hog enetic protein (bBMP)with same weight was buried in the muscles of the behind le gs of the mouse.The place which bBMP was buried in was examined with ultrasonog r aphy,X-ray and histological section at 3,5,7,14,21,28 day after the operat ion re spectively.Results No new bone was found by ul trasonography and X-ray at 3,5,7,14 day and the signs of new bone appeared at 21 ,28 day.By histological method a large number of mesenchymal cells were seen i n the muscles in which bBMP was buried at 3,5,7 day,and the formation of new ca rtilage and the new bone at 14 day,then the normal bone developed at 21,28 day .Conclusion The results of B-mood ultrasound an d X-ray and histological section were almost the same.So it seems feasible that B-mood ultrasound is regarded as a new method to determine the bone induction ab ility of bBMP.%目的 探讨一种测定骨形成蛋白(BoneMorphogeneticProtein,BMP)成骨诱导活性的新方法。方法 将自行提取的牛BMP(bBMP)埋入小白鼠后腿肌袋内,分别于术后3、5、7、14、21、28d行B超、X射线照相和组织形态学观察。结果 B超3、5、7d与对照组织同,均为正常组织回声,14、21、28d有软骨和骨组织回声。X射线照相3、5、7、14d无骨组织影象,21、28d有明显的新生骨征象。组织形态学观察3、5、7d有大量间充质细胞在BMP植入部位聚集,14d有新生软骨和新生骨组织组成,21、28d可见大量的新生的成熟骨组织。结论 B超、X射线照相和组织形态学观察结果在时间上有很强的一致性,证明B超作为一种新的BMP成骨诱导活性的检测手段是切实可行的。
