与感觉相关的ASIC3、TRPV1基因敲除小鼠繁殖性能的观察与痛阈的比较

摘要

Objective To investigate the reproductive performance and compare pain threshold of ASIC3-/-and TRPV1-/-mice with C57BL/6 mice.Method 12 ASIC3-/-mice,12 TRPV1-/-mice and 12 C57BL/6 mice were chosen and monogamously mated.There are 6 male and 6 female mice in each group,with age of 8-10 weeks.The pregnancy rate,the litter size,and the weaning rate of the ASIC3-/-mice and TRPV1-/-mice were analyzed.The pain threshold among C57BL/6mice,ASIC3-/-mice and TRPV1-/-mice were compared.Result The pregnancy rate,the litter size,and the weaning rate of the ASIC3-/-mice is slower than that of the TRPV1-/-mice.Comparing with G57BL/6 mice,the tactile threshold of ASIC3-/-mice was significantly higher (P ＜0.001) and the thermal threshold of TRPV1-/-mice was significantly higher (P ＜0.001).Conclusion The reproductive performance of the ASIC3-/-mice is weaker than that of the TRPV1-/-mice.The tactile sensitivity of the ASIC3-/-mice decreased.And the thermal sensitivity of the TRPV1-/-mice decreased.%目的 观察酸敏感离子通道亚基3(ASIC3,又名ACCN3)基因敲除ASIC3-/-小鼠、香草酸瞬时受体亚型Ⅰ(TRPV1)基因敲除TRPV1-/-小鼠的繁殖性能,并对其基础痛阈值进行测定和比较.方法 选用成熟的未交配过的ASIC3-/-和TRPV1-/-纯合子小鼠分别进行繁殖交配,统计两种小鼠的妊娠率、窝产仔数、离乳率、头胎产仔数等,并对两种小鼠的基础痛阈值进行测定比较,对照组为同源野生C57BL/6小鼠.结果 ASIC3-/-小鼠的妊娠率、窝产仔数、离乳率较TRPV1-/-小鼠偏低,而TRPV1-/-小鼠妊娠率、窝产仔数、离乳率相对稳定；ASIC3-/-组的基础机械痛阈值与C57BL/6对照组相比显著升高(P ＜0.001),而热痛阈值没有明显改变；TRPV1-/-组的基础热痛阈值与C57 BL/6对照组显著升高(P＜0.001).结论 ASIC3-/-小鼠的繁殖能力较TRPV1-/-小鼠弱；ASIC3-/-小鼠的机械痛敏感性下降,TRPV1-/-小鼠的热痛敏感性下降.
