碳纳米管对单核巨噬细胞作用的基因表达谱研究

             

摘要

Objective:Using Affymetrix Genechip, we had studied the possible mechanism of p-MWCNTs and tau-MWCNTs on RAW264. 7 macrophages. Methods: Changes of gene expression in RAW264. 7 macrophages, after treatment with p-MWCNTs and tau-MWCNTs, were examined by the cDNA microarray in order to study the molecular mechanism. Real time RT-PCR was conducted to verify the differential expression of some genes. Results: After the RAW 264. 7 macrophages were exposed to the p-MWCNTs for 24h, compared with the control, there were 5738 genes showed differential expression) On the other hand, after the RAW 264. 7 macrophages were exposed to the tau-MWCNTs for 24h, there were 2 567 genes showed differential expression. According to the Gene Ontology, these genes were involved in cell cycle, apoptosis, stress and inflammation, cytoskeleton and so on. Conclusions: The p-MWCNTs inhibit the proliferation of the RAW 264. 7 macrophages by regulating the genes expression, including cell cycle, apoptosis, stress, inflammation and so on. There are reduced differential expression genes on the RAW 264. 7 macrophages after treatment with the tau-MWCNTs.%目的:采用Affmetrix公司的基因组芯片,研究原始的多壁碳纳米管(pristine multi-walled carbon nanotubes,p-MWCNTs)与牛磺酸修饰的多壁碳纳米管(taurine functionalized multi-walled carbon nanotubes,tau-MWCNTs)对小鼠的单核巨噬细胞RAW264.7细胞的作用机制.方法:通过对差异表达基因的生物学功能予以分类,从分子水平上探讨p-MWCNTs与tau-MWCNTs对RAW264.7细胞的作用机制,并以实时荧光定量聚合酶链反应来验证研究结果.结果:与对照组相比,p-MWCNTs处理RAW264.7细胞24 h后,RAW264.7细胞表达显著改变的基因共5 738个;而经tau-MWCNTs处理RAW264.7细胞24 h后,RAW264.7细胞表达显著改变的基因共2 567个,这些基因的功能涉及细胞周期、细胞凋亡、应激反应、炎性反应与细胞骨架等.结论:P-MWCNTs抑制RAW264.7细胞的生长是通过调控一些与细胞周期、细胞凋亡、应激反应及炎性反应相关的基因而起作用.而tau-MWCNTs处理RAW264.7细胞后发生变化的基因数量显著减少.
