左归丸对糖皮质激素所致骨质疏松大鼠血清中E2、PTH含量的影响

摘要

Objective To reveal the mechanism that how the Zuogui Pill treat glucocorticoid induced osteoporosis. Methods; Virgin Wistar rats were selected and randomly divided into three groups ( n = 12-15 per group) , including control group, model group and Zuogui pill group. The rats were injected dexamethasone for eight weeks to make the model of osteoporosis and the Zuogui Pill were administered intragastrically to the rats of Zuogui Pill group for eight weeks. We analyzed the effect of Zuogui Pill on tibia bone by measuring the relative parameters of the histomorphometric slides of the undecalcified tibia. And the content of E2 and PTH in serum of rats were detected before and after the use of Zuogui Pill. Results;We found that TBV% and TFS% in model group decreased significantly, while TRS% increased significantly. And the content of PTH in serum increased significantly in model group, while E2 decreased significantly. After being treated by prescriptions of Zuogui pill, We found that TBV% and TFS% increased significantly in Zuogui pill group, no changes of TRS% has been observed, while it has the tendency to decrease. The content of PTH in serum increased significantly in Zuogui pill group ,while E2 increased significantly. Conclusion;Zuogui Pill can treat glucocorticoid-induced osteoporosis of rats. One of the mechanism is that Zuogui Pill can decrease serum PTH level and increase the content of E2 in serum.%目的:探讨左归丸对糖皮质激素性骨质疏松症的治疗作用及机理.方法:将42只雌性Wistar大鼠随机分为3组:空白对照组12只,模型组15只,左归丸组15只.造模给药8周后,采用骨组织形态计量学方法对不脱钙骨切片进行形态计量.并采用ELISA 法检测大鼠外周血血清中E2、PTH的含量.结果:与空白对照组相比,模型组大鼠胫骨TBV%及TFS%显著降低,TRS%显著增高;且大鼠血清中PTH含量显著增高,E2含量则显著降低.而左归丸组大鼠TBV%则显著高于模型组,TFS%亦显著增高,TRS%虽无差异,但有明显降低趋势;且血清中PTH含量显著降低,E2含量较之模型组显著增高.结论:左归丸可使糖皮质激素所致骨质疏松大鼠血清中PTH的水平下调,同时上调E2水平,使其紊乱状态基本恢复正常,这可能是其治疗糖皮质激素所致大鼠骨质疏松症的机理之一.
