基于腺性膀胱炎模型大鼠探讨八正散对IL-6、IL-17、TGF-β1表达的影响

         

摘要

Objective To study the effects of Bazheng San on the expressions of IL-6, IL-17, and TGF-β1 in bladder of rats suffering from glandular cystitis (GC).Methods The GC rats were divided into the blank group, the model group, the control group, high, medium and low dose of Bazhensan group. Rats in the control group were given HCPT(3mg/kg•d) by intravesical administration, and rats in high, medium and low dose Bazhengsan group were given with Bazhengsan by intragastric administration 2 times a day for 2 weeks with 0.84g/mL, 0.42g/mL, 0.21g/mL respectively. Then the pathological changes of bladder tissue was observed and IL-6, IL-17, TGF-β1 expression in bladder of rat were detected by immunohistochemistry.Results Compared with that of the model group, the pathological changes of bladder mucosa lamina propria were improved in the control group and high dose Bazhensan group including inflammatory cells infiltration, interstitial edema, focal necrosis, brunn nest and gland metaplasia. Comparing that of the blank group, IL-6, IL-17 and TGF-β1 expressions were increased obviously in the model group. And these cytokines level were decreased significantly in the control group and Bazheng San-duct high-dose group. The data also showed that IL-6 and TGF-β1 level in Bazheng San-duct medium-dose group were decreased (P<0.05).Conclusion Bazheng San may reduce the expression level of IL-6, IL-17, TGF-β1 in CG rats.%目的:探讨八正散对腺性膀胱炎病理模型大鼠相关细胞因子的影响机制。方法以腺性膀胱炎模型大鼠为研究对象,随机分为空白组、模型组、对照组和八正散的不同剂量(高、中、低)组。对照组给予羟基喜树碱每天3mg/kg膀胱灌注,八正散高、中、低剂量组分别给予同体积八正散液(生药浓度分别为0.84g/mL、0.42g/mL、0.21g/mL),每日2次,灌注/灌胃连续2周。HE染色观察膀胱病理变化,免疫组织化学法观察膀胱组织中的白细胞介素6(interleukin6,IL-6)、白细胞介素17(interleukin17,IL17)、转化生长因子β1(Transforming growth factor-β1,TGF-β1)的表达。结果对照组及八正散高剂量组大鼠膀胱黏膜固有层炎细胞浸润、间质水肿、局灶性坏死和黏膜固有层内Brunn巢及腺上皮化生等较模型组明显减少。与空白组比较,模型组的IL-6、IL-17和TGF-β1的含量显著升高,差异有统计学意义(P<0.01);与模型组比较,对照组及高剂量八正散组IL-6、IL-17和TGF-β1的含量显著降低,差异有统计学意义(P<0.05)。中剂量组IL-6、TGF-β1的含量显著降低,差异有统计学意义(P<0.05)。结论八正散可以有效改善腺性膀胱炎模型大鼠膀胱病变程度,推测与下调IL-6、IL-17和TGF-β1含量有关。
