长期培养大鼠海马脑片CA1区基质细胞诱导因子和CXCR4蛋白的免疫活性变化

             

摘要

目的:研究基质细胞诱导因子-1α(SDF-1α)和CXCR4蛋白在长期培养的海马脑片CA1区的表达变化.方法:运用免疫组织化学法检测SDF-1α和CXCR4在大鼠海马脑片CA1区的表达变化.结果:培养1~5周海马脑片CA1区均有SDF-1α和CXCR4的蛋白表达,阳性细胞的胞质呈棕黄色染色.培养1周时,CXCR4蛋白的表达水平较高,随着培养时间的延长,表达呈下降的趋势,培养至5周的海马脑片CA1区CXCR4蛋白表达水平和培养1周相比差异有统计学意义.培养1周的海马脑片CA1区SDF-1α的表达也处于较高水平,培养至2~3周时有所下降,但与培养1周相比无差异,培养至4~5周时其表达则下降.结论:SDF-1α和CXCR4蛋白在大鼠海马脑片CA1的表达变化具有时空性差异,提示SDF-1/CXCR4生物轴参与了海马脑片在体外的生长,并可能介导了CA1区神经元细胞的分化.%Objective-. To investigate the expressional changes of stromal cell-derived factor-lα (SDF-1α) and CXCR4 proteins in CA1 area during the long-term hippocampal organotypic tissue culture (HOTC) in rats. Methods-. Immuno-histochemical analysis was used to detect SDF-la and CXCR4 expression in CA1 area of hippocampus slice. Results-. The immunohistochemical reactions of SDF-1α and CXCR4 were positive in CA1 area of hippocampal slice with the cell membrane and cytoplasm stained from cultured 1 week to cultured 5 weeks. The expression of CXCR4 was higher at 1 week when it was compared with the cultured 5 weeks, and then its expression was descending trend. The expression of SDF-1α protein was high at cultured 1 week, while its expression was decreased gradually from cultured 2 weeks to 5 weeks. Conclusion-. The protein expressions of SDF-1α and CXCR4 in long-time HOTCs of rats are of spatial-temporal dependent features, which implys that the SDF-1/CXCR4 axis take part in the development of HOTC, and may be related to cell differentiation of CA1 area.
