冠心病患者缺氧诱导因子-1α与血红素氧化酶-1的相关性研究

摘要

Objective To investigate the correlation between hypoxia inducible factor - 1α( HIF - 1α ) and heme oxygenase - 1( HO - 1 ) in patients with coronary artery disease. Methods A total of 95 patients with coronary artery disease ( 30 acute myocardial infarction,30 unstable angina pectoris, and 30 stable angina pectoris ) and 30 control subjects with normal coronary artery were collected according to coronary angiogram. The protein expressions of HIF - 1α and HO - 1 in monocytes and lymphocytes were detected by Western blotting, and then the relationship between HIF - 1α and HO - 1 was investigated. Results Compared with the control group , the expression of HIF - 1α and HO - 1 in AMl and UAP group increased significantly, but increased slightly in SAP group. There was a positive correlation between the expression of HIF - 1α and HO - 1 in the total level ( r =0. 76 ) as well as in each case group. Conclusion The HIF - 1α/HO - 1 pathway is activated in the development of coronary artery disease , and HO - 1 is induced by HIF - 1 to participate in the cardioprotection.%目的 探讨冠心病患者外周血单个核细胞缺氧诱导因子-1α(HIF-1α)与血红素氧化酶-1(HO-1)的相关性.方法 选择经冠脉造影确诊的冠心病患者为病例组,按照临床表现分为急性心肌梗死组(AMI组)、不稳定型心绞痛组(UAP组)和稳定型心绞痛组(SAP组);选择冠脉造影未发现冠脉病变的患者为对照组.利用Western blot检测入选对象外周血单个核细胞HIF-1α和HO-1的蛋白表达水平,相关分析方法分析两者表达水平的相关性.结果 与对照组相比,AMI和UAP组HO-1和HIF-1α蛋白表达均明显增高,SAP组患者增高不明显.总体水平上,HIF-1α与HO-1呈正相关(r=0.76);在AMI、UAP、SAP组HIF-1α与HO-1表达水平也呈正相关.结论 冠心病发生、发展过程中HIF-1α/HO-1通路被激活,HIF-1α可能通过诱导HO-1蛋白表达参与心肌保护作用.
