糖尿病合并冠心病患者血红素氧化酶-1基因多态性的表达及其意义

     

摘要

目的 研究糖尿病患者血红素氧化酶-1(HO-1)基因多态性及HO-1表达水平与冠心病易感性的关系.方法 选择经冠状动脉造影证实为冠心病或排除冠心病的糖尿病患者分别为62例和59例,采用聚合酶链反应(PCR)检测HO-1启动子区GT重复[(GT)n]多态性并进行基因分型,用蛋白质印迹法(Western blot)测HO-1的表达水平.结果 (GT)n重复次数在14~39之间,将(GT)n重复序列分为S(n<27)和L(n≥27)两种等位基因,SS、SL、LL 3种基因型,冠心病组的S等位基因明显少于对照组(P<0.01),SS基因型亦明显少于对照组(P<0.05);冠心病组HO-1蛋白表达明显低于对照组(P<0.01) 结论 糖尿病人群中,HO-1的基因多态性与冠心病易感相关,拥有长重复序列(L等位基因)者,其HO-1表达水平明显减低,为冠心病易感人群.%Objective To assess the relationship of coronary artery disease and heme oxygenase-1 (HO-1) gene promoter polymorphism, expression level of HO-1 in the patients with diabetes mellitus.Methods 62 patients with diabetes mellitus who had positive results in coronary angiography (CAG) were defined as CHD group.And 59 subjects who had negative results in CAG were divided as control group.HO-1 gene promoter polymorphism and genotyping were determined using polymerase chain reaction.HO-1 was extracted and measured using western blot analysis.Results (GT)n polymorphism was found in HO-1 gene promoter with 14-39.The distribution of numbers of (GT)n repeats was divided into two subclasses: class S included shorter (n<21) repeats, and class L included longer (n≥27) repeats.The subjects were then classified as having an SS, SL, or LL genotypes.The frequency of S allele was less in the CHD group compared with the control group (P < 0.01), and SS genotypes was also less than that of the control group (P < 0.01).The levels of HO-1 were significantly lower in the CHD group than that of the control group (P < 0.01).Conclusion HO-1 gene promoter polymorphism is correlated with coronary heart disease in the patients with diabetes mellitus.Longer (GT)n repeat allele in HO-1 promoter and the lower levels of HO-1 take part in the emotivity of CHD in patients with diabetes.
