β-catenin、RIP K4、CDK8在套细胞淋巴瘤中的表达及临床意义

Objective To detect the expressions ofβ⁃catenin, RIPK4, CDK8 and assess the relationship between the expres⁃sions of above proteins in mantle cell lymphoma( MCL) with their clinicopathological characteristics and prognosis. Methods Strept Avidin⁃Biotin Complex immunohistochemistry was used to detect the expression ofβ⁃catenin, RIPK4 and CDK8 in 30 samples of MCL and 16 samples of lymphadenitis. All MCL patients were followed⁃up. The expressions of β⁃catenin, RIPK4 and CDK8 in MCL and lymphadentis tissues were compared. The correlation of above proteins and the relationship between the expression of them in MCL with their clinicopathological characteristics and prognosis were analyzed. Results Positive expression rates of β⁃catenin, RIPK4 and CDK8 in MCL tissues were 55�3%, 63�3% and 60�0%, which were higher than those of lymphadenitis samples( P<0�05) . There was a positive correlation among the expression of β⁃catenin, RIPK4 and CDK8( r=0�600,r=0�675,r=0�367,P<0�05) . The expression of β⁃catenin, RIPK4 and CDK8 had no significant correlation with the gender, age, MIPI score, the level of β2⁃microglobulin, B symptoms and clinical stage(P>0�05). The median overall survival (OS) of MCL patients was 21 months. MIPI score of high risk group, the positive expression of CDK8 were independent predictors of OS. Conclusion There were high expression rate ofβ⁃catenin, RIPK4 and CDK8 proteins in MCL. There was a positive correlation among the expression ofβ⁃catenin, RIPK4 and CDK8. The positive expression of CDK8 indicated a poor prognosis of MCL.%目的：探讨β⁃catenin、RIPK4及CDK8在套细胞淋巴瘤（ MCL）组织中的表达及其与临床病理特征、预后之间的关系。方法采用免疫组化SABC法检测30例MCL组织和16例淋巴结炎性病变组织中β⁃catenin、RIPK4及CDK8的表达情况，并对MCL患者进行生存随访。分析β⁃catenin、RIPK4及CDK8表达的相关性及其与MCL临床病理特征、预后之间的关系。结果 MCL组织中β⁃catenin、RIPK4及CDK8的阳性表达率分别为55�3％、63�3％和60�0％，均高于淋巴结炎性病变组织（P＜0�05）；MCL 组织中β⁃catenin、RIPK4及 CDK8的表达任两者均呈正相关（r＝0�600，r＝0�675，r＝0�367，P＜0�05）；β⁃catenin、RIPK4及CDK8的表达与性别、年龄、MIPI评分、β2微球蛋白、B症状、Ann Arbor分期均无关（ P＞0�05）。30例MCL患者的中位总生存期（ OS）为21个月，多因素分析结果显示MIPI评分高危组、CDK8阳性表达是影响OS的独立不良预后因素。结论β⁃catenin、RIPK4及 CDK8在 MCL 中高表达；β⁃catenin、RIPK4及 CDK8的表达呈正相关；CDK8阳性表达提示预后不佳。
