为明确山西省晋中市侵染番茄的病毒种类,采用双链RNA(double-stranded RNA,dsRNA)技术和非序列依赖PCR扩增(sequence-independent amplification,SIA)方法对感病番茄进行分子鉴定.序列测定及分析发现,具有花叶、卷曲症状的番茄为烟草花叶病毒(Tobacco mosaic virus,TMV)所侵染.进一步克隆TMV番茄分离物(TMV-SXFQ)的全基因组序列,得到TMV-SXFQ(GenBank登录号为JX993906)全长为6 394 bp,含4个开放阅读框(open reading frame,ORF).序列比对分析表明,TMV-SXFQ与烟草花叶病毒属其他分离物的同源性为86.3% ~ 99.6%;系统进化分析表明,TMV-SXFQ与韩国分离物IM聚为一簇、亲缘关系最近.%To identify the virus pathogen inducing tomato (Solanum lycopersicum L.) mosaic and curl disease at Jinzhong City of Shanxi Province,the technology of double-stranded RNA (dsRNA) and sequenceindependent amplification (SIA) were used.Results of SIA and sequencing showed that there was Tobacco mosaic virus (TMV) in affected tomato leaves.Further cloning the whole genome sequence of TMV (TMV-SXFQ),the TMV-SXFQ was gained (GenBank No.JX993906),which was 6 394 hp in length and presented a typical Tobamovirus genome organization with 40RFs.Sequence comparative analysis showed that the homology of TMV-SXFQ and Tobacco mosaic virus and other isolates was 86.3%-99.6%.Phylogenetic analysis suggested that TMV-SXFQ isolates had closeer ties consanguinity with IM strain from South Korea and these 2 isolates formed an independent branch.
