GC法同时测定维药芜菁子中4种异硫氰酸酯类化合物的含量

         

摘要

OBJECTIVE: To establish the method for the content determination of isothiocyanate compounds in the seeds of Brassica rapa. METHODS: Isothiocyanates was extracted from the seeds of B. rapa by steam distillation method. Using isooctanol as internal standard, its components and contents were analyzed by GC. RESULTS: There were 4 kinds of isothiocyanates in the seeds of Brassica rapa, such as phenylhexyl isothiocyanat (14.20% ), text-butyisothiocyanate (0.47%), isopropyl isothiocyanate (0.29%),allyl isothiocyanate(5.10%). The linear ranges of 4 compounds were 10.06—50.30 μg(r=0.999 6),0.25—1.26 μg(r= 0.999 5),2.83—14.15 μg(r=0.998 9) and 3.34—16.72 μg(r=0.998 6), respectively. Average recovery rates were 99.2%, 100.2% , 97.8% and 98.2%, and RSDs were 1.3%, 1.9%, 2.3% and 2.1% (n=9), respectively. CONCLUSION: The method is simple, sensitive, specific and reproducible. It can be used for quality control of the seeds of B. rapa.%目的:建立同时测定维药芜菁子中异硫氰酸酯类化合物含量的方法.方法:采用水蒸气蒸馏法提取芜菁子中的异硫氰酸酯,以异辛醇为内标物,利用气相色谱法分析异硫氰酸酯类化合物的组成并测定其含量.结果:芜菁子中含有4种异硫氰酸酯类化合物,分别为异硫氰酸苯乙酯(14.20%)、异硫氰酸叔丁酯(0.47%)、异硫氰酸异丙酯(0.29%)和异硫氰酸烯丙酯(5.10%).4种化合物的进样量线性范围依次为10.06~50.30 μg(r=0.999 6)、0.25~1.26 μg(r=0.999 5)、2.83~14.15 μg(r=0.998 9)、3.34~16.72 μg(r=0.998 6);平均加样回收率分别为99.2%、100.2%、97.8%、98.2%,RSD分别为1.3%、1.9%、2.3%、2.1%(n均为9).结论:本方法简便、灵敏、专属性强、重复性好,可用于芜菁子的质量控制.
