哈蟆油延缓衰老的作用机制研究

         

摘要

OBJECTIVE: To study the anti-aging effect of Rana temporaria chensiensis on aging model mice. METHODS: Model mice were divided into blank control group, aging/hypoimmunity model group and R. temporaria chensiensis high-dose,medium-dose and low-dose groups(0.4,0.2,0.1 g·kg-1). The aging mice model was induced by hypodermic injection of D-galactose. The contents of MDA, NO and GSH and the activity of XOD and MPO were determined, the hypoimmunity mice model was induced by X ray irradiation and the content of fatty acid in blood of hypoimmunity mice were determined by GC method. RESULTS: Compared with blank control group, the content of MDA and the activity of XOD in aging model group increased significantly while the contents of NO and GSH and the activity of MPO decreased significantly(P<0.01 or P<0.05). Compared with the aging model group, the content of MDA and the activity of XOD in R.temporaia chensinensis high-dose, medium-dose and low-dose groups decreased significantly, while the activity of MPO increased significantly(P<0.01 or P<0.05).The activity of NO in R.temporaia chensinensis low-dose group increased significantly(P<0.01).Compared with the blank control group,the content of EPA/AA, EPA, fatty acids, unsaturated fatty acid,PUFA in model group decreased significantly(P<0.05 or P<0.01); Compared with the hypoimmunity model group,the content of toad oil fatty acids, DHA, EPA, n-3PUFA,unsaturated fatty acid, PUFA in R. temporaia chensinensis low-dose, medium-dose and low-dose groups increased significantly (P< 0.01 or P<0.05) , EPA/AA in R. temporaia chensinensis low-dose and medium-dose groups decreased significantly(P<0.01) ,the content of DHA/AA,saturated fatty acid in R. temporaia chensinensis low-dose group decreased significantly(P<0.01 or P<0.05).CONCLUSION: It shows that R. temporaria chensiensis has the significant antioxidant effect and can increase the content of unsaturated fatty acid of hypoimmunity model mice, which may be the mechanism of it' s anti-aging effect.%目的:研究哈蟆油抗衰老的作用机制.方法:实验分为空白对照、衰老/免疫低下模型与哈蟆油高、中、低剂量(0.4、0.2、0.1 g·kg-1)组.采用皮下注射D-半乳糖复制小鼠衰老模型,测定肾脏丙二醛(MDA)、一氧化氮(NO)、还原型谷胱甘肽(GSH)的含量和髓过氧化物酶(MPO)、黄嘌呤氧化酶(XOD)的活性;采用深部X射线照射复制小鼠免疫低下模型,以气相色谱法测定小鼠血液中脂肪酸的含量.结果:与空白对照组比较,衰老模型组小鼠肾脏MDA含量显著上升,XOD活性显著升高,NO、GSH含量显著下降,MPO活性显著下降(P<0.01或P<0.05);与衰老模型组比较,哈蟆油高、中、低剂量组小鼠MDA含量显著降低,XOD活性显著减弱,MPO活性显著增强(P<0.01或P<0.05),哈蟆油低剂量组NO含量显著升高(P<0.01).与空白对照组比较,免疫低下模型组二十碳五烯酸(EPA)/二十碳四烯酸(AA)、EPA、脂肪酸总量、单不饱和脂肪酸、多不饱和脂肪酸(PUFA)含量显著降低(P<0.05或P<0.01);与免疫低下模型组比较,哈蟆油高、中、低剂量组二十二碳六烯酸(DHA)、EPA、脂肪酸总量、n-3系PUFA、单不饱和脂肪酸、PUFA含量显著增加(P<0.05或P<0.01),哈蟆油中、低剂量组EPA/AA显著降低(P<0.01),蛤蟆油低剂量组DHA/AA、饱和脂肪酸含量显著减少(P<0.05或P<0.01).结论:哈蟆油具有显著的抗氧化作用,可以增加免疫力低下小鼠体内不饱和脂肪酸含量,这些可能是其延缓衰老的机制之一.
