荧光分光光度法检测维库溴胺的血药浓度

         

摘要

Objective To establish a rapid and sensitive fluorescence spectrophotometric method for determining the plasma concentration of vecuronium. Methods The plasma samples were subsequently extracted with trichloromethane after basification by carbonate, and detected at the excitation wavelength of 550 nm and the emission wavelength of 590 nm by fluorescence spectrophotometry. Results The linear range for vecuronium was Y= 1.235 +4.01 C, r=0. 995 8. The lowest limit of determination was 0. 05 μg/mL. The average recovery rate was more than 81.28%,the intra-day and inter-day variations were both less than 8%. Conclusion The method is simple, rapid and sensitive with good reproducibility, which is suitable for the determination vecuronium in human plasma.%目的 建立快速、灵敏的荧光分光光度法,用于测定人血浆中维库溴胺的质量浓度.方法 血浆样品用碳酸盐碱化、三氯甲烷萃取后,用荧光分光光度计在激发波长550 nm、发射波长590 nm处进行检测.结果 维库溴胺的线性回归方程为Y(荧光强度)=1.235+4.01 C,r=0.995 8.维库溴胺最低检测质量浓度为0.05 μg/Ml,平均回收率在81.28%以上,日内和日间变异小于8%.结论 该法简单、快速、灵敏、重现性好,是一种检测人血浆中维库溴胺浓度的有效方法.
