高效液相色谱法测定复方消渴滴丸中葛根素含量

摘要

Objective To establish an HPLC method for the determination of assay and of puerarin in Fufangxiaoke Pills. Methods The chromatographic conditions were C18 columne quipped with a C18 column(250 mm ×4. 6 mm,5 μm), with the mobile phase of methanol : water(28 : 72),the flow rate was l.0mL/min and the detective wavelength was 250 nm, the injection volumn was 10 μL. Results The puerarin curves were linear over the concentration range of 83.68-836.8 μg/mL( RSD =0. 999 8), and the average recovery was 99.08% for three different levels of the added amount of puerarin, RSD = 1. 09% ( n = 9). Conclusion The method is simple and reproducible. It can be used as the preparation of quality control methods.%目的 建立复方消渴滴丸中葛根素的含量测定方法.方法 采用高效液相色谱法,色谱柱为C18柱(200 mm×4.6 mm,5μm),流动相为甲醇-水(28∶72),流速1.0 mL/min,柱温30℃,紫外检测波长250 nm,进样体积10 μL.结果 葛根素质量浓度在83.68～836.8 μg/mL内与峰面积线性关系良好,r=0.999 8(n=7).高、中、低3种不同质量浓度溶液的平均回收率为99.08％,RSD=1.09％(n=9).结论 该方法简便易行、重复性好,可作为复方消渴滴丸的质量控制方法.
