反相高效液相色谱法同时测定香菊胶囊中绿原酸和咖啡酸含量

         

摘要

目的:建立同时测定香菊胶囊中绿原酸和咖啡酸含量的反相高效液相色谱法。方法色谱柱采用Agilent Zorbax SB-C18柱(250 mm ×4.6 mm,5μm),流动相为乙腈-甲醇-0.2%磷酸溶液(线性梯度洗脱),检测波长328 nm,流速1.0 mL/min,进样量20μL,柱温30℃。结果绿原酸和咖啡酸进样量分别在2.034×10-2~5.085×10-1μg ( r=0.9999,n=7)和2.218×10-2~5.545×10-1μg ( r=0.9999,n=7)时与峰面积线性关系良好,加样回收率分别为98.83%和98.72%( RSD分别为0.86%,0.88%,n=6)。结论该法操作快速、简便、准确、结果可靠,可用于香菊胶囊的质量控制。%Objective To establish an RP-HPLC method for simultaneous content determination of chlorogenic acid and caffeic acid in Xiangju Capsules. Methods The Agilent Zobax SB-C18 colume ( 250 mm × 4. 6 mm, 5 μm ) was used; the mobile phase was acetoni-trile-methanol-0. 2%H3PO4 by gradient elution and the detection wavelength was 328 nm;the flow rate was 1. 0 mL/min, the injec-tion volume was 20 μL, and the column temperature was 30 ℃. Results The linear response of chlorogenic acid ranged from 2. 034 × 10-2-5. 085 × 10-1μg ( r=0. 999 9,n=7 ) , and the linear response of caffeic acid ranged from 2. 218 × 10-2-5. 545 × 10-1μg ( r=0. 999 9,n=7 );the average recovery rate of chlorogenic acid was 98. 83% ( RSD=0. 86%,n=6 ) , and the average recovery of caffeic acid was 98. 72% ( RSD=0. 88%,n=6 ) . Conclusion The method is convenient, quick and accurate, and can be used for the quality control of Xiangju Capsules.
