目的 观察幽门螺杆菌(H.pylori,Hp)作用于食管癌细胞后细胞色素氧化酶I(cytochrome oxidase I,COX I)mRNA及其蛋白表达的变化,探讨Hp在肿瘤发生发展中可能的作用机制.方法 将Hp与食管癌细胞分别作用4、8、24 h.收集细胞,应用Real-Time PCR和Western blot法检测各时间点COX I mRNA及其蛋白表达.结果 Hp与食管癌细胞共培养4、8、24 h后,COX I mRNA表达量逐渐减少,4 h的表达量显著低于对照组,其后下降速度减慢,24 h后表达量不再有明显改变.COX I蛋白表达呈现相似的趋势.结论 Hp与食管癌细胞共培养后,可导致线粒体编码基因COX I mRNA及其蛋白表达障碍,从而影响线粒体功能.%Objective To study the effects of H. pylori (Hp) on the expressions of COX I mRNA and protein in esophageal carcinoma cells, and explore the potential role of Hp in carcinogenesis. Methods After ECa-109 cells were treated by Hp for 4, 8, 24 h respectively, the expression of COX I mRNA and protein was detected by Real-Time PCR and Western blot. Results After ECa-109 cells were treated by Hp for 4, 8, 24 h respectively, the expression of COX J mRNA decreased gradually, after exposure to Hp for 4 h, the expression of COX I significantly lower than control group, than the decreasing speed slowed When treated with Hp for 24 h, the expression have no significant changes. The expression of COX .1 protein revealed the same trend. Conclusion Hp can affect mitochondrial function by down regulating the expression of mitochondria] encoding cytochrome oxidase I gene.
