PIK3CA及GST-π在人骨肉瘤中的表达及其临床意义

             

摘要

Objective To detect the expression of phosphatidylinositol 3-kinase (PIK3CA) and glutathione-S transferase-π (GST-π) in osteosarcoma tissues and to explore its effect on the multidrug resistance of osteosarcoma.Methods Thirty-three cases of pathological tissue specimens of osteosarcoma from Renmin Hospital of Wuhan University from January 2009 to June 2017 were collected.The expression of PIK3CA and GST-π of 33 cases of osteosarcoma specimens was detected by immunohistochemistry (Envision method) and Western blot method.The differences between the chemotherapy-resistant group (7 cases) and the chemotherapy-effective group (26 cases) were compared.The results were analyzed combined with clinicopathological factors.Results The average optical density of PIK3CA in the chemotherapy-resistant group and chemotherapy-effective group was (11.089±2.147) and (6.938±2.065) respectively,and there was a significant difference between the two groups (P < 0.05);the average optical density of GST-π in the chemotherapy-resistant group and chemotherapy-effective group was (15.871±2.678) and (7.693±2.443),and there was a significant difference between the two groups (P < 0.05).The average optical density of PIK3CA and GST-π by Western blot method had significant differences between the two groups (P < 0.05).The expression of PIK3CA and GST-π in osteosarcoma was correlated (r =0.68,P < 0.01).Conclusion PIK3CA and GST-π are over expressed in osteosarcoma chemotherapy-resistant group,and the expression of PIK3CA and GST-π is positively correlated with each other,which may be one of the mechanisms of drug resistance in osteosarcoma.%目的 检测磷脂酰肌醇3激酶(PIK3CA)及谷胱甘肽-S-转移酶π(GST-π)在骨肉瘤组织中的表达,探讨其对骨肉瘤多药耐药的影响.方法 收集武汉大学人民医院2009年1月~2017年6月33例骨肉瘤病理组织标本,应用免疫组化方法(Envision法)及Western blot法测定PIK3CA、GST-π在骨肉瘤标本中的表达.比较化疗耐药组(7例)和化疗有效组(26例)中表达的差异,并结合临床病理因素进行分析.结果 免疫组化检测化疗耐药组和化疗有效组的PIK3CA表达的平均吸光度值分别为(11.089±2.147)、(6.938±2.065),差异有统计学意义(P<0.05);化疗耐药组和化疗有效组GST-π表达的平均吸光度值分别为(15.871±2.678)、(7.693±2.443),差异有统计学意义(P<0.05).Western blot法检测两组中PIK3CA和GST-π的蛋白表达,差异均有统计学意义(P<0.05).PIK3CA和GST-π在骨肉瘤组织中的蛋白表达具有相关性(r=0.68,P< 0.01).结论 PIK3CA及GST-π在骨肉瘤化疗耐药组中高表达,且两者表达呈正相关,其可能是骨肉瘤耐药的发生机制之一.
