活体荧光成像仪用于小鼠细菌感染的监测

     

摘要

Objective:To establish a mouse model of sepsis and execute noninvasive imaging for mouse of bacterial infection.Methods: In vivo fluorescence imager was applied to achieve vitro imaging for luminous bacteria, and Pseudomonas aeruginosa (typical Gram-negative bacteria) was injected through tail vein to induce septicopyemia for establishing mouse model. And then, The bacterial infection of the mice were analyzed by using vivo imaging technique. Results: Bacterial bioluminescence readings which could reflect the activity of bacterial luciferase were positively correlated with bacterial colony forming units (CFU) which were confirmed through routine counting. The survival rate of infected mice depended on dose and time of bacterial infection of various concentration.Conclusion: In vivo fluorescence imager can detect bacterial bioluminescence readings and it can be used to monitor the changes of bacterial propagation for infected mice.%目的:建立一种脓毒血症小鼠模型,对细菌感染情况进行非损伤性成像.方法:采用活体荧光成像仪对发光细菌进行体外成像,通过小鼠尾静脉注射铜绿假单胞菌(典型的革兰氏阴性细菌)诱导脓毒血症建立小鼠模型,利用活体成像分析小鼠体内的细菌感染情况.结果:反映细菌荧光素酶活性的细菌生物发光读数与通过常规计数确定的细菌菌落形成单位(CFU)数呈正相关.细菌感染小鼠的存活率对不同浓度水平的细菌感染有剂量和时间依赖性.结论:活体荧光成像仪可检测细菌生物发光读数,可以应用于监测小鼠感染细菌增殖的变化.
