大肠癌细胞对化疗药物的敏感性与p53基因突变的相关性研究

         

摘要

目的研究大肠癌细胞对化疗药物的敏感性与p53基因突变的相关性.方法用MTT比色法检测20位大肠癌患者的癌细胞对化疗药物的敏感性,并用PCR-SSCP技术分析其癌细胞的p53基因突变.结果大肠癌细胞对5-Fu[IR=(54.7±14.1)%]比对MMC[IR=(43.9±10.8)%]及ADM[IR=(43.9±9.4)%]更敏感(P<0.05),但对DDP有耐药性[IR=(25.9±12.8)%,P<0.01].p53基因的突变率为45%(9/20).癌细胞对MMC和ADM的敏感性与p53基因突变呈正相关(rmmc=0.589,rabm=0.624,P<0.005).结论化疗药物MMC和ADM对大肠癌的作用依赖于p53基因的状态,而大肠癌对5-Fu和DDP的敏感性却与p53基因突变无关.%Objective: To investigate the correlation between the sensitivity of colorectal carcinoma cells to chemotherapeutic medicines and mutation of p53 gene. Methods:20 patients with colorectal carcinoma were observed,utilizing MTT colorimetry to determine colorectal carcinoma cells' sensitivity to chemotherapeutic medicines and PCR- SSCP technique to analyze mutation of p53 gene. Results :The colorectal carcinoma cells were more sensitive to 5 -Fu [IR=(54.7 + 14.1)%] than to MMC [IR=(43.9 + 10.8)%, P<0.05] and ADM [IR=(43.9±9.4) %, P < 0. 05 ], and resistant to DDP [ IR = (25.9 + 12. 8) %, P < 0.01 ]. The mutation rate of p53 gene was 45% (9/20). A positive correlation was found between sensitivities of MMC and ADM and mutation of p53 gene(rmmc=0.589, radm = 0.624, P < 0. 005 ). Conclusions: The effects of chemotherapeutic medicines such as MMC and ADM on colorectal carcinoma depend on the status of p53 gene. However,there is no correlation between sensitivities to 5 - Fu and DDP and mutations of p53 gene.
