HPLC法测定生脉注射液中特征成分莽草酸

         

摘要

Identification of shikimic acid in Shengmai injection was confirmed by liquid chromatography-mass spectrometry,To establish an HPLC method for determining Shikimic acid in Shengmai injection. The liquid chromatography-mass spectrometry determination confirmed that the target peak was Shikimic acid by comparing the molecular ion peak weight (m/z:173.08 [M-H]-) with Shikimic acid standard substance.The analysis was performed on a Synergi Hydro-RP 80A (250 mm×4.6 mm,4μm), mobile phase was 20 mmol/L KH2PO4 (tune pH to 2.5 with H3PO4), flow rate was 0.6 mL/min, detection wavelength at 214 nm. It was a good linear relationship of Shikimic acid content among 0.02 to 0.64μg, r=0.9998. The RSD of precision and repeatability were 0.15% and 1.49% respectly. The average recovery was 99.2%, RSD=1.56%. The study identified the existence of Shikimic acid in Shengmai injection for the first time. And the component derived from Schisandra chinensis. The content determination method was accurate and reliable. It could be used for the quality control of Schisandra chinensis in Shengmai injection.%采用液质联用法对生脉注射液中莽草酸成分进行确认,建立高效液相色谱法测定生脉注射液中莽草酸含量.采用液质联用法测定目标峰的分子离子峰m/z:173.08[M-H]-,与莽草酸对照品对比确认目标峰为莽草酸成分;色谱柱采用Synergi Hydro-RP 80A(250 mm×4.6 mm,4μm),以20 mmol/L的KH2PO4(H3PO4调pH至2.5)溶液为流动相,流量为0.6 mL/min,检测波长214 nm;莽草酸在0.02~0.64μg范围内呈良好的线性关系,r=0.9998,精密度、重复性试验RSD分别为0.15%和1.49%,平均加样回收率为99.2%,RSD=1.56%.该研究首次确认生脉注射液中含有莽草酸成分,且该成分来源于五味子;含量测定方法准确、可靠,可作为生脉注射液中五味子成分质量控制检测方法.
