缺氧模型用于扩管药耳蜗效应评定的研究

         

摘要

目的:探讨缺氧模型用于评定扩管药耳蜗效应的可行性,并确立适宜的低氧浓度及评定参数,以期为临床用药提供筛选方法。方法:分别在豚鼠7%,5%低氧供模型上,以ABR为指标,观察具有耳蜗扩管作用的丹参、川芎嗪和罂粟碱的耳蜗扩管效应是否得以体现。结果:与对照组相比,7%低氧供30min后,3种扩管药的耳蜗扩管效应均通过ABR阈移的显著降低得以体现,波I潜伏期及振幅的变化在各组间无显著差异;5%低氧供30min后,仅丹参和罂粟碱的耳蜗扩管作用得以证实。结论:低氧下听觉电生理反应模型可用于评价扩管药在耳蜗的效应,其适宜低氧浓度为7%,ABR阈移可用作评定药效时的观察指标。%Objective: To explore the possibility to evaluate the cochlear effect of vasodilator by the hypoxia model. Methods:theeffects of salvia mitiorribizae,ligustrazine and papaverine which had been testified to be improve the cochlear microcirculation on theABR were be o bserved in the guinea pig hypoxic modd(7%O2 or 5%O2). Results: Conpared with the control group, after animalswere subjected to hypoxia of 7% oxygen for 30 minutes,the cochlear effect of three vasodilators were be confirmed by the marked reduce of the ABR threshold shift,the latency and the amplitude of wave I didn't appear and differen~ among groups. after ventilatedwith hypoxic gas of 5% oxygen for 30 minute,only the salvia mitiorribizae and the papaverine's cochlear effect were be verified. Conclusion: the model of cochlear electrophysiology during hypoxia could be utilized to evaluate the cochlear effect of vasodilator, the appropriate hypoxia gas is 7% oxygen,the threshold shift of ABR could be used as indexes.
