穴位注射川芎嗪抑制氧诱导小鼠视网膜新生血管形成的实验研究

             

摘要

目的 探讨穴位注射川芎嗪抑制氧诱导小鼠视网膜新生血管形成的作用.方法 将60只C57BL/6J小鼠的健康幼鼠(出生后7d),随机分为正常对照组、模型对照组(A组)、穴位注射组(B组)、全身用药组(C组),每组15只.除正常对照组外,其余3组于75%高氧环境中饲养5d,建立氧诱导的视网膜新生血管病变幼鼠模型.B组、C组分别予穴位注射和鼠尾静脉注射川芎嗪稀释液,A组予鼠尾静脉注射等体积的生理盐水.采用双抗体夹心法测定血清血管内皮生长因子(vascular endothelial growth factor,VEGF)含量,光学显微镜下观察视网膜新生血管内皮细胞核数目.结果 1.血清VEGF含量:正常对照组(9.28±1.26)μg/L、B组(13.35±2.41)μg/L、C组(12.37±3.11) μg/L,均低于A组(18.31±4.33) μg/L,差异有统计学意义(P<0.01,P<0.05);B组与C组比较,差异无统计学意义(P>0.05).2.视网膜新生血管内皮细胞核数:A、B、C组均可见视网膜新生血管芽,发生率为100%.正常对照组及A、B、C组的新生血管细胞核数分别为6.56±3.41、130.83±41.63、68.39±32.07、59.39+21.78.A组明显高于正常对照组,差异有统计学意义(P<0.01);B组、C组均低于A组,差异有统计学意义(P<0.05);B组与C组比较,差异无统计学意义(P>0.05).结论 川芎嗪能减少血管增生性视网膜病变小鼠的血清VEGF含量,抑制氧诱导小鼠视网膜新生血管形成.川芎嗪穴位注射可有效作用于视网膜新生血管病变.%OBJECTIVE To investigate the inhibition effects of acupuncture point injection of Chuanxiongzine on oxygen-induced retinal neovascularization in mice. METHODS Seven-day old C57BL/6J 60 mice were randomly divided into normal control group, model control group (A), model acupuncture point injection group (B) and model tail vein injection group(C). Except for the normal control group, other groups were exposed to 75% oxygen for 5 days and then to room air. Group B was given with acupuncture point injection of Chuanxiongzine and group C was given with tail vein injection of Chuanxiongzine and group A was treated with the equal bulk of saline by intraperitoneal injection. Serum VEGF was detected by double antibody methods. The numbers of neovascular en-dothelium were counted and compared under optical microscope. RESULTS Sixty mice were included. 1.The levels of serum VEGF of group B (13.35±2.41) μg/L and group C (12.37±3.1) μg/L were significantly lower than that of group A( 18.31±4.33) μg/L(P<0.01); The level of serum VEGF of group A was significantly higher than that of normal control group (9.28±1.26) μg/L (P<0 05); The levels of serum VEGF between group B with group C had no difference ( P>0.05); 2. New vessel buds were observed in all groups. The incidence of new vessels in model groups was 100%. The numbers of new vessel buds in the normal control group, group A, group B, and group C were 6.56± 3.41, 130.83±41.63, 68.39±32.07 and 59.39+21.78 respectively. Compared with group A, retinal neovascularization in group B and group C were reduced (P<0 05); The new vessel buds of group A and B had no difference (P>0.05 ).CONCLUSIONS Chuanxiongzine could decrease the levels of VEGF and the amount of new vessel buds in oxygen-induced retinal neovascularization mice model; Acupuncture point injecting Chuanxiongzine could effectively inhibit retinal neovascularization.
