分子荧光法快速测定饲料中的金霉素

     

摘要

In order to develop the rapid determination of chlortetracycline content in feed,the samples were deter-mined by molecular fluorescence spectrometry after extraction with ultrasonic technology.The results showed that the opti-mal excitation wavelength was 350 nm for chlortetracycline hydrochloride,emission wavelength was 421 nm,the conversion time was 60 min,transformation temperature was 40 ℃,pH was 11.50,ultrasonic temperature was 60 ℃,ultrasonic power was 160 W and the material liquid ratio was 1∶50.The regression equation of work curve was Y=798.32+21.208X,the lin-ear correlation coefficient (r) equalled 0.9996,detection limit was 0.544 ng/mL,quantitative limit was 1.813 ng/mL.The standard addition recovery rate was between 92.00% and 103.83%,and relative standard deviation (RSD) was less than 1%(n=11).%为快速测定饲料中金霉素含量,将实验样品采用超声波浸提后,用分子荧光法快速测定。结果表明,盐酸金霉素的最佳分析条件为:激发波长350 nm,发射波长421 nm,转化时间60 min,转化温度40℃,pH 11.50,超声温度60℃,超声功率160 W,料液比1∶50。工作曲线的回归方程为Y=798.32X+21.208,相关系数r=0.9996,检出限为0.544 ng/mL,定量限1.813 ng/mL;样品加标回收率为92.00%~103.83%,相对标准偏差(RSD)小于1%(n=11)。
