超声波辅助消解-紫外可见分光光度法测定大肉姜中的硒

     

摘要

O-phenylendiamine is used as chelating agent,with ultrasonic wave assisting the digestion of Zingiber officinale,complexation and extraction process of Se-o-phenylenediamine are determined.The content of selenium in Zingiber officinale is determined by UV visible spectrophotometry.The results show that the wavelength is 334 nm.This method has a good linearship of selenium content in the range of 0~2.08 μg/mL,and the correlation coefficient is 0.9999.The best conditions for samples' digestion are:digestion system of HNO3-HClO4,volume ratio of 4∶1,mixed acid dosage of 18 mL,ultrasonicassisted digestion time of 30 min,water bath temperature of 50 ℃,the selenium content in Zingiber officinale of 4.4425 μg/g,the adding standard recovery rate of 97.79%~102.02%.The result of digestion is efficacious and satisfactory.%以邻苯二胺为络合显色剂,超声波辅助大肉姜的消解及硒与邻苯二胺的络合、萃取过程,利用紫外可见分光光度法测定大肉姜中的硒含量.结果表明:体系测定波长为334 nm,硒在0~2.08 μg/mL含量范围内有良好的线性关系,相关系数为0.9999,样品的最佳消解条件:消解体系为HNO3-HClO4,体积比为4∶1,用量为18 mL的混酸,超声辅助消解时间为30 min,水浴温度为50 ℃,测得大肉姜中的硒含量为4.4425 μg/g,加标回收率为97.79%~102.02%,消解效果好,结果令人满意.
