ε-聚赖氨酸生产菌的硫酸二乙酯诱变及其发酵培养基优化

     

摘要

Streptomyces albulus FQ-23 is mutagenesised by diethyl sulfate in this study.After flat screening and shake flask screening,a high-yielding strain with an ε-poly-L-lysine yield of 0.976 g/L is obtained.It is named as Streptomyces albulus FQF-3.It could be stably hereditary and used for follow-up study.Four major components:glucose,(NH4)2SO4,corn steep liquor and fish meal are optimized by single factor experiment and response surface methodology.The final optimization results are (NH4)2SO4 of 9.95 g/L,fish meal of 12.52 g/L,corn steep liquor of 7.79 g/L,glucose of 49.70 g/L,and the remaining component of M3G medium is the most suitable medium.Under this medium,the production of ε-poly-L-lysine is 1.373 g/L,which is 1.53 times higher than that of FQ-23.%对出发茵白色链霉菌FQ-23进行硫酸二乙酯诱变,经过平板初筛和摇瓶复筛后,获得1株ε-聚赖氨酸产量为0.976 g/L的高产菌株,并将其命名为白色链霉菌FQF-3.可稳定遗传,并用于后续研究.然后通过单因素与响应面分析方法对4大主要成分:葡萄糖、硫酸铵、玉米浆和鱼粉进行了优化,最终优化结果为:硫酸铵、鱼粉、玉米浆、葡萄糖的含量分别为9.95,12.52,7.79,49.70 g/L.再加上M3G培养基的剩余成分即为最适培养基.在该培养基下,ε-聚赖氨酸产量为1.373 g/L,是出发茵产量的1.53倍.
