假单胞菌基因敲除方法研究进展

摘要

Gene knockout is a very popular genetic engineering technology in recent years.Pseudomonas is a common bacterium,which is applied very widely.Therefore,gene knockout in Pseudomonas is very significant.We review the successful case of gene knockout in Pseudomonas,and compare two kinds of gene knockout systems in the following aspects:transformation method of key step in homologous recombination,choice of gene knockout method,and choice of the length of homologous arm,etc.%基因敲除是近年来十分热门的基因工程技术,假单胞菌(Pseudomonas)作为一种常见细菌,应用十分广泛,因此,对假单胞菌进行基因敲除改造有着十分重要的意义.总结了假单胞菌基因敲除技术成功案例,并对2种基因敲除系统进行了比较,包括同源重组关键步骤的转化方式、基因敲除方法的选择、同源臂长度的选择等.
