浓缩菜籽磷脂制备磷脂酰胆碱工艺研究

             

摘要

In this paper,the method of extraction with ethanol was used to make up the phosphatidylcholine from concentrated rapeseed phospholipid.Then the phosphatidylcholine content of the product was analysed with high performance liquid chromatography. The effects of four factors included solid–liquid ratio,ethanol concentration,extraction time and temperature on the ethanol extraction result were discussed with the method of single factor and orthogonal experiments. It is concluded that the optimal parameters for the process of ethanol extraction were as follows:solid–liquid ratio 1∶7,temperature 50℃,concentration of ethanol 95%,extraction time 45min. Under this condition,the phosphatidylcholine percentage content of the obtained product was 42.92%,and the yield of phosphatidylcholine was 61.03%.%  以浓缩菜籽磷脂为原料,采用乙醇萃取法制取菜籽磷脂酰胆碱浓缩物,应用高效液相色谱法检测分析产品中磷脂酰胆碱含量.重点探讨乙醇萃取过程中料液比、乙醇浓度、萃取时间和萃取温度等工艺条件对提取效果影响,采用单因素实验和正交实验优化工艺参数,得出乙醇萃取制备菜籽磷脂酰胆碱最优工艺参数为:料液比1∶7、萃取温度50℃、乙醇浓度95%、萃取时间为45 min;在此条件下得率为61.03%,磷脂酰胆碱含量为42.92%.
