气相色谱法测定饲料中金刚烷胺药物的含量

摘要

实验建立了气相色谱法检测饲料中金刚烷胺含量的方法,饲料中的金刚烷胺用甲醇:三氯乙酸=1:1提取,取上层提取液经MCX阳离子交换柱净化,用氨水+异丙醇+甲醇=5+25+70混合液5 ml洗脱,经氮吹浓缩,浓缩物在碱性条件下用三氯甲烷溶剂复溶,取有机层进行气相色谱仪-FID测定,外标法定量,方法检出限达到1.0 mg/kg,方法回收率达到76.7% ~98.3%,相对标准偏差小于5%.%The determination method of amantadine by gas chromatography in feed was established.Amantadine in feed was extracted with 1:1 ratio of methanol to trichloroacetic acid,The upper extract was purified by MCX cation exchange column, which eluted by 5 ml mixed liquor(composed of ammonia 5,isopropanol 25 and methanol 70),it was concentrated by nitrogen concentration,the concentrate is redissolve by trichloromethane in alkaline conditions,and the organic layer was determined by the gas chromatograph FID,By means of the external standard method,the detection limit reached 1.0 mg/kg,the recovery rate of the method reached 76.7%-98.3%,and the relative standard deviation was less than 5%.
