HPLC 法测定清热暗疮丸中绿原酸的含量

摘要

Objective To develop an HPLC method for the determination of chlorogenic acid in heat acne pills.Methods Thermo U-3000 HPLC,ChromeleonTM7,and Thermo DAD-3000 Detector were adopted.The column was Thermo C18 (250 mm ×4.6 mm,5 μm). The mobile phase was acetonitrile -0.4%phosphoric acid solution(14∶86).The flow rate was 1.0 mL·min -1 .The column tempera-ture was set at 25℃ and UV detection wavelength was 327 nm.Injection volume was 10 μL.Results The linearity range of chloro-genic acid was 2.556 ~255.6 mg·L -1 (r =0.999 99).The average recovery of adding sample was 99.8%(n =9).The examination limit was 0.051 mg·L -1 .The RSD(n =5)of measurement precision test,reproducibility test and stability were 0.039%,0.69% and 0.60%,respectively.Concluslons This method is simple,stable with good separation efficiency and accurate result,which is suitable for the determination of chlorogenic acid in heat acne pills.%目的：建立 HPLC 法测定清热暗疮丸中绿原酸的含量。方法Thermo U-3000液相色谱系统；ChromeleonTM7工作站；Thermo DAD-3000检测器；Thermo C18（250 mm ×4．6 mm，5μm）色谱柱；流动相：乙腈—0．4％磷酸溶液（14∶86）；流速：1．0 mL ·min －1；柱温：25℃；紫外检测波长327 nm；进样量为10μL。结果绿原酸在2．556～255．6 mg·L －1的范围内呈良好的线性关系（r ＝0．99999）；绿原酸的平均加样回收率为99．8％（n ＝9）；检测限为0．051 mg·L －1；精密度试验 RSD ＝0．039％；重现性实验 RSD ＝0．69％；稳定性实验 RSD ＝0．60％。结论该方法简便、稳定，分离效果好，结果准确，适用于清热暗疮丸中绿原酸的质量控制。
