Objective To study the effects of extracellular matrix on the proliferation and migration of HK-2 cells. Methods The MDCK extracellular matrix was used as the basis for the culture of HK-2 cells and randomly divided into normal control group and extracellular matrix group. The condition of cell proliferation and migration was observed with acid phosphatase and cell scratches; the expression of integrin α3 and α6 protein was analyzed by immunofluorescence and Western blot. Results Compared with normal group,MDCK extracellular matrix significantly promoted the proliferation and migration of HK-2 cells in vitro,and increased the expression of integrin α3 and α6(P < 0. 05). Conclution MDCK extracellular matrix could promote the proliferation and migration of HK-2 cells cultured in vitro.%目的 研究MDCK 细胞外基质对HK-2 细胞的增殖和迁移的影响.方法 用氨水制备MDCK 细胞外间质,并且作为HK-2 细胞培养基础,随机分为正常对照组和细胞外基质组.用酸性磷酸酶、细胞划痕实验观察细胞的增殖和迁移状况;细胞免疫荧光和Westen blot 分析整合素α3 和α6蛋白的表达变化.结果 与正常对照组比较,MDCK 细胞外基质能明显促进HK-2 细胞的体外增殖和迁移,整合素α3和α6 表达增加(P < 0. 05).结论 MDCK 细胞外基质可促进体外培养的HK-2 细胞的增殖和迁移.
