白芍总苷对糖尿病大鼠肾组织TLR2、TLR4/NF-κB信号通路调节的影响

         

摘要

目的 研究白芍总苷(TGP)对糖尿病大鼠肾组织中Toll样受体2(TLR2)、Toll样受体4(TLR4)、核转录因子-κB(NF-κB-p-p65)表达的影响.方法 将链脲佐菌素(STZ)诱导的糖尿病大鼠模型随机分为模型组、TGP组(50、100、200 mg/kg·d),正常大鼠为对照组,8周后免疫组化法检测肾组织中TLR2、TLR4、NF-κB-p-p65的表达.结果 模型组大鼠尿白蛋白排泄率(AER)明显高于对照组(P<0.01),TGP(50、100、200 mg/kg·d)给药8周后大鼠AER水平明显低于模型组(P<0.05,P<0.01).模型组大鼠肾组织中TLR2、TLR4与NF-κB-p-p65表达明显高于对照组(P<0.01),TGP(50、100、200 mg/kg·d)给药8周肾小球TLR2表达与模型组相比差异无显著性,肾小管-间质TLR2表达明显低于模型组(P<0.01),TGP(50、100、200 mg/kg·d)给药8周肾小球与肾小管-间质TLR4、NF-κB-p-p65表达明显低于模型组(P<0.05,P<0.01).结论 TGP可显著改善糖尿病大鼠早期肾脏损害,其机制与部分抑制糖尿病大鼠肾组织中TLR2、TLR4/NF-κB信号通路有关.%Objective To investigate the effect of total glucosides of paeony ( TGP) on the expression of toll-like receptor 2(TLR2) , TLR4 and NF-κB-p-p65 in the kidney from experimental diabetic rat.Methods Diabetes rats were induced with streptozotocin (STZ) ,and TGP (50,100,200 mg/kg·d ) was orally administered once a day for 8 weeks. Normal rats served as control group The expressions of TLR2, TLR4 and NF-κB-p-p65 were determined by immunohistochemical method in the kidney. Results Urinary albumin excretion rate ( AER) in model group was markedly obviously higher than that in the control group (P <0. 01) , AER in model group was markedly attenuated than that in TGP ( 50,100 and 200 mg/kg· d ) treatment Compared with control rats , the expressions of TLR2, TLR4 and NF-κB-p-p65 were significantly increased in the kidney. TGP (50,100 and 200 mg/kg· d) treatment could markedly reduce TLR2 expression in the tubulo -interstitium, TLR4 and NF-κB-p-p65 expression in the glo- meruli and tubulo-interstitium. Conclusion TGP treatment can ameliorate early renal injury by inhibiting the over -expression of TLR2,4/ NF-κB signal pathway in the kidney from diabetic rats.u0000b
