转OjDREB基因提高烟草耐盐能力的研究

     

摘要

利用农杆菌介导法将沿阶草OjDREB基因转入烟草植株中.通过RT-PCR检测,获得转OjDREB基因的烟草植株.以400 mmol/L NaC1处理转基因烟草植株和野生型对照10 d后,结果显示:转基因烟草体内叶绿素含量和SOD酶活性是野生型对照的16.98倍和1.91倍;体内的MDA含量和电解质渗透率分别是野生型对照的59.23%和55.07%,说明过量表达OjDREB基因可以提高烟草的耐盐能力.%Ophiopogon japonicus DREB gene OjDREB was introduced into tobacco plant by Agrobacterium tumefaciens mediated transformation leaf disc. The expression of exogenous OjDREB gene in transgenic tobacco was identified by RT-PCR. Transgenic and wild-type plants were irrigated with 400 mmol/L NaCl for 10 days. Results showed that chlorophyll content and SOD activity of transgenic tobacco were significantly higher than that of wild-type tobacco, by 16. 98 times and 1. 91 times respectively. While MDA content and relative leakage was lower than that of wild-type, 59. 23% and 55. 07% of the wild-type respectively. It was shown that overexpression of OjDREB gene improved salt tolerance in tobacco plant, which can be used in molecular breeding.
