Chimeric genes using cDNA of the glucose-2 transporter (GLUT2) in antisense orientation and transgenic non-human animals expressing said chimeric genes for use in the study of diabetes mellitus and the development of therapeutic approaches to the disease.Concerns a chimeric gene using cDNA (complementary DNA) of the glucose-2 transporter (GLUT2) in reversed orientation, directed by a promoter or promoter fusion.It also concerns a chimeric gene obtained by the fusion of GLUT2 cDNA in antisense orientation with the insulin gene promoter and a chimeric gene obtained by fusion of GLUT2 cDNA in antisense orientation with the P-enolpyruvate carboxykinase promoter (PEPCK).It also concerns an expression vector for the expression of antisense GLUT2 RNA in any cell type, notably a pRIPantiGT2 and pInsantiGT2 plasmid vector, a pPCKantiGT2 plasmid vector, a vRIPantiGT2 and vInsantiGT2 retroviral vector and a pPCKantiGT2 retroviral vector.It also concerns transgenic non-human animals which express the aforementioned chimeric gene and a cell type originating in said transgenic non-human animals.Applies to the development of therapeutic approaches to diabetes mellitus.
