国产新华联屋尘螨点刺液生物活性的研究

     

摘要

目的 探讨国产新华联屋尘螨点刺液的体内和体外生物活性.方法 收集60例单纯尘螨过敏的哮喘患者血清,制备屋尘螨血清池,采用放射过敏原免疫吸附(RAST)抑制试验方法测定新华联屋尘螨点刺液体外生物活性并与德国阿罗格屋尘螨点刺液和丹麦ALK屋尘螨点刺液进行比较;选择90例疑诊过敏性疾病患者,采用新华联屋尘螨点刺液和阿罗格屋尘螨点刺液进行皮肤点刺试验,比较其体内生物活性.采用聚丙烯酰胺凝胶电泳法比较不同屋尘螨点刺液蛋白组分有无差异;酶联免疫吸附法(ELISA)测定屋尘螨点刺液主要致敏蛋白Der p1的含量.结果 RAST抑制试验显示3批次的新华联屋尘螨点刺液均能抑制标准屋尘螨抗原与IgE的特异性结合,其体外生物活性与阿罗格和ALK屋尘螨点刺液相似;皮肤点刺试验显示新华联屋尘螨点刺液的体内生物活性与阿罗格屋尘螨点刺液相似;电泳结果提示3批次的新华联屋尘螨点刺液蛋白组分相同,与阿罗格和ALK屋尘螨点刺液组分相似;ELISA法检测发现3批次的新华联屋尘螨点刺液主要致敏蛋白Der p1含量相同,低于ALK屋尘螨点刺液,但高于阿罗格屋尘螨点刺液.结论 国产新华联屋尘螨点刺液具有较强的体内和体外生物活性,可用于屋尘螨过敏的诊断.%Objective To investigate the bioactivity of commercial Chinese Derrnatophagoides Pteronyssinus (Dp)skin prick solution in vivo and in vitro. Methods The sera from 60 patients with dust-mite allergic asthma were collected to prepare an allergic sera pool. Radioallergosorbent test(RAST)inhibition was performed to test bioactivity of Xinhualian, Allergopharma and ALK skin prick solution respectively in vitro. Skin prick tests were performed on 90 patients suspected of having allergic diseases with both Xinhualian and Allergopharma skin prick solution to compare the bioactivity in vivo. Polyacrylamide gel electrophoresis(PAGF) was used to compare the protein ingredients of those three kinds of Dp skin prick solutions. Fnzyme-linked immunosorbent assay( ELISA) was used to measure the content of major allergenic protein Der pl of Dp skin prick solutions. Results RAST revealed that Xinhualian Dp skin prick solution could inhibit specific IgE's binding ability to standard Dp allergen in vitro ,which was similar to Allergopharma and ALK skin prick solution. The skin tests showed that Xinhualian and Allergopharma Dp skin prick solution had the same bioactivity in vivo. PAGE and FLISA results demonstrated that the protein ingredients and the content of Der pl of the three batches of Dp solutions were the same. The content of Der pl of Xinhualian Dp skin prick solution was lower than that of ALK, but higher than that of Allergopharma. Conclusion Xinhualian Dp skin prick solution has a good bioactivity both in vivo and in vitro. The skin prick solution is very stable and reliable in clinical diagnosis of Dp allergy.
