STAT3在子宫内膜癌组织中的表达及与ER、PR相关性研究

摘要

目的 探讨信号转导与转录活化因子3(STAT3)与雌激素受体(ER)、孕激素受体(PR)在子宫内膜癌中的表达及其相关性.方法 采用免疫组织化学技术检测50例子宫内膜癌标本,10例正常子宫内膜标本的STAT3、ER、PR的表达.结果 在子宫内膜癌组织中STAT3、ER、PR的阳性率分别为64%、62%、50%,在对照组子宫内膜组织中STAT3、ER、PR的阳性表达率分别为20%,100%、100%,差异均有统计学意义(P值分别为0.027、0.047、0.010).癌组织中STAT3的表达与病理分级、手术-病理分期、淋巴结转移有相关性(P值分别为0.006、0.026、0.027),癌组织中ER的表达均与病理分级、手术-病理分期、淋巴结转移有相关性(P值分别为0.002、0.007、0.030).PR的表达与病理分级、手术-病理分期有关(P值分别为0.004、0.030),而与淋巴结转移无相关性(P值为0.061).子宫内膜癌组织中STAT3与PR呈正相关(rs =0.417,P=0.003),但与ER无相关关系(rs =0.271,P=0.057).结论 STAT3为子宫内膜癌的治疗提供新的分子治疗靶点,子宫内膜癌组织中STAT3与PR的表达呈正相关,而与ER无关,STAT3表达可能被孕激素调控,而这一激活过程与ER无关.%Objective To investigate the expression of signal transducer and activator of transcription3 (STAT3) in endometrial carcinoma tissues and its correlation with estrogen receptor (ER) and progastrone receptor (PR). Methods Immunohis-tochemical staining was used to detect the expression of STAT3,ER and PR in 50 cases of endometrial carcinoma tissues and 10 normal endometrial tissues. Results The positive expression rate of STAT3, ER and PR was 64% ,62% ,50% in endometrial carcinoma tissues respectively,significantly different from that in normal endometrial tissues with the positive rate being 20% , 100% and 100% respectively (P<0. 05) . The expression of STAT3 and ER was closely related to histological grade, FIGO stage,lymph node metastasis in endometrial carcinoma. The expression of PR was associated with histological grade and FIGO stage but not with lymph node metastasis in endometrial carcinoma. The expression of all the three proteins had no relationship with age. Positive correlation was found between the expression of STAT3 and PR (rs=0. 417,P = 0. 003) but not between the expression of STAT3 and ER (rs= 0. 271 ,P = 0. 057) in endometrial carcinoma. Conclusion The detection for STAT3 ,ER and PR expression may help evaluate biological behaviors of endometrial carcinoma and the prognosis of patients, which may provide a new molecular therapeutic target for endometrial carcinoma. The expression of STAT3 is positively correlated with the expression of PR but not with ER in endometrial carcinoma.
