目的:从穿龙薯蓣根茎中分离纯化内生真菌,筛选能产生皂苷类成分的功能菌株.方法:通过组织分离法从穿龙薯蓣根茎中分离内生真菌,利用高效液相色谱法(HPLC)和液相色谱-质谱法(HPLC-MS)对分离得到的内生真菌发酵产物进行分析和检测.结果:共分离纯化内生真菌50株,HPLC检测发现内生真菌C12能产生与薯蓣皂苷元具有相同紫外吸收的结构相近化合物,经HPLC-MS分析鉴定该成分为△3,5-去氧替告皂苷元.结论:穿龙薯蓣内生真菌资源丰富,其中内生真菌C12具有产生皂苷类成分的功能.%Objective:To isolate and screen endophytic fungi producing diosgenin or other aglycone from the fresh rhizome of Dioscorea nipponica Makino.Methods:The endophytic fungi from Dioscorea nipponica Makino was isolated by tissue block method,the metabolites of diosgenin was detected by HPLC and LC-MS.Results:A total of 50 strains of the endophytic fungi were isolated,which C12 strain in the fermentation broth contained similar structure compounds of Diosgenin,which was identified as △3,5-Deoxyneotigogenin by HPLC-MS data analysis.Conclusion:There are a large number of endophytic fungi in Dioscorea nipponica Makino,and the endophytic fungi have the ability to produce the same or similar components as the host plants.
