不同NAA 和6-BA 对牡丹胚性愈伤组织诱导的影响

         

摘要

以牡丹品种'凤丹白'愈伤组织为材料,研究不同NAA和6-BA对其胚性愈伤组织诱导效果,以及其抗氧化酶(POD、SOD、CAf)活性和可溶性蛋白变化的影响.结果表明:不同处理中以添加 NAA0.2 mg/L+6-BA 0.5 mg/L处理的胚性愈伤组织诱导效果最好,该处理下愈伤组织于诱导培养的第20天胚性细胞形成,第25天出现大量胚性细胞,第30天有少量早期球形胚,在胚性愈伤组织形成过程中存在非胚性细胞与胚性细胞、球形胚共存的现象.在此处理下POD、CAf活性在20~25 d呈显著上升趋势,明显优于其它处理,SOD活性在整个处理期间比较稳定,且一直保持较高水平.可溶性蛋白的含量在胚性细胞出现和分化的过程中一直保持较高水平,对形成胚性愈伤组织有促进作用.%To elucidate the effects of phytohormone on the activity of antioxidase and the changes of solu-ble protein in the somatic embryogenesis,the somatic embryogenic callus of Paeonia suffruticosa'Fengdanbai' was propagated on MS medium supplemented with different concentrations of NAA and 6-BA,and then the ac-tivities of POD,SOD and CAT were determined.The results showed that the hormone combination of 0.2 mg/L NAA and 0.5 mg/L 6-BA was the best for embryogenic callus induction.Under this treatment the embryonic cells began to form at 20 d,and appeared in a large number at 25 d,and a bit of early globular embryos arose at 30 d.In the process of embryogenic callus formation,non-embryonic and embryonic cells,and early globular embryos coexisted.The activity of POD and CAT exhibited a remarkable rising trend and higher than that in other treatments at 20-25 d,moreover,the activity of SOD was stable and maintained at a high level throughout the experiment.The soluble protein content maintained a high level in the appearing and differentiating process of embryonic cells,which was beneficial to the formation of embryonic callus.
