春兰杂交种子非共生萌发和快速繁殖

         

摘要

Cymbidium goeringii hybrid seeds were sown aseptically to induce the root stock.The results demonstrated that when the medium was MS + 0.1 mg/L BA+ 0.1 mg/L NAA+ 5 g/L LH + 1 g/L AC,the seed germination rate reached 66.67 % , and adding LH could obviously improve the seed germination rate and promote the growth of the root stock.The 8~9-month-old capsule was comparatively ideal in germination rate.When the differential medium was WPM+ 1.0 mg/L 6-BA+ 5 g/L LH+ 1 g/L AC,the multiplication coefficient of the root stock reached 2.7 and the bud differentiation rate was up to 76.7% ,in which the suitable culture cycle was 35 ~ 45 d.When the rooting medium was 1/2 MS +1.0 mg/L NAA+ 2.5 g/L LH,the rooting rate was 95 % and the survival rate of transplanted plantlets was 83 % .%春兰杂交种子非共生萌发即无菌播种诱导根状茎试验表明:培养基为MS+BA 0.1 mg/L+NAA0.1 mg/L+LH 5g/L+AC 1g/L时,其萌发率达到66.67%,添加水解乳蛋白可明显提高种子发芽率并促进根状茎生长,果龄8~9个月的蒴果播种萌发率较为理想;根状茎增殖分化培养基为WPM+6-BA 1.0 mg/L+LH5 mg/L+AC 1g/L时,其根状茎增殖倍数为2.7,芽苗分化率最大达76.7%,培养适宜周期为35~45 d;生根培养基为1/2 MS+NAA 1.0 mg/L+LH 2.5 g/L时,生根率达到95%,移栽成活率为83%.
