不同方法处理对椰子花粉扫描电镜观察结果的影响

         

摘要

分别用冻干、常温干燥器干燥、36℃烘干、液氮冷冻、-80℃冷冻等5种方法对海南高种椰子花粉的电子扫描显微镜观察样品进行预处理.结果表明:以冻干法的效果最好;常温干燥器干燥与36℃烘干法制备的花粉样品在扫描电镜下观察呈现塌陷、变形状;-80℃冷冻与液氮冷冻处理的雄花外壁呈褐色,花药、花丝潮湿,电镜观察视野中的杂质、碎片较多.%Five methods (freeze- drying, silica- gel drying at room- temperature, 36 ℃ drying, liquid nitrogen freezing, -80 ℃ freezing) were used to pretreat the male flowers of coconut, and then scanning electron microscope (SEM) was used to observe the morphology of the pollen. The results showed that the freeze - drying pretreatment had the best effect for the observation of coconut pollen, and the exine ornamentations of the pollens pretreated by freeze - drying were clear. While some of the pollens pretreated by 36 ℃ drying or silica - gel drying at room - temperature were of collapse or distortion. The male flowers pretreated by liquid nitrogen freezing or -80 ℃ freezing were wet and brown, their anthers and filaments were wet, and there were many impurities and fragments in the perspective of SEM.
