以黄粉虫幼虫的水溶性蛋白为原料,利用碱性蛋白酶和胰蛋白酶水解制备抗氧化肽,并对其水解工艺进行优化.以·OH清除率为指标,在单因素的基础上设计3因素(加酶量、pH和酶解温度)3水平的二次正交旋转组合试验.结果表明,双酶水解的最佳工艺条件为:m(料)∶V(液)=1∶20、水解时间2 h、加酶量17.36 mg/g(碱性蛋白酶和胰蛋白酶酶活比为1∶1)、pH8.0、温度46.5℃.在此条件下制得的多肽具有较强的还原力,对DPPH·和·OH有较强清除作用.%In order to obtain the antioxidant peptides, water-dissolved protein of Tenebrio molitor L.was hydrolyzed by alkali protease and trypsase.Conditions of enzymatic reaction were determined and optimized.By measuring · OH radical scavenging rates, quadratic regression orthogonal rotary method was designed basing on single factor test, including three factors (dosage of protease, pH, and temperature) and three levels.The result showed that the optimal hydrolytic conditions were, reaction time 2 hours, ratio of solid to liquid 1∶ 20 (m∶ V), dosage of protease 17.36 mg/g (ratio of protease energy 1∶ 1), pH 8.0 and temperature 46.5 ℃.The obtained peptides showed high reducing activity, · OH radical scavenging rates and DPPH · radical scavenging rates.
