Solid-phase synthesis of hydantoins as caspase inhibitors

摘要

Apoptosis is a critical process in a wide variety of biological systems,including normal cell turnover,the immune system,embryonic development and metamorphosis.Inappropriate apoptosis occurs in many human pathologies,including neurodegenerative diseases.Designs based on apopain structure have provided potent peptidic and pseudopeptidic inhibitors,but with physical-chemistry and pharmacokinetic characteristics that limit their use to the intravenous treatment of acute episodes,of some diseases.It is therefore necessary to search for new structures and scaffolds that reduce these limitations.
