Purification and properties of 3- #alpha# -hydroxysteroid dehydrogease and 3-Keto-5- #beta# -steroid- #DELTA# ~4-dehydrogenase

The level of the concentrations of total bile acids (TBA) in serum is an important index of the diagnosis of hepatic diseases, the conventional measuring methods of which are so complicated, time-consuming, and expensive, requiring expensive instrumentation and are thus unsuitable for routine clinical use. A new method using an enzyme kit to determine the amount of TBA in serum appeared recently, which is easier to standardize and less expensive. The enzymatic method is also sensitive and can given better analytical performance, so is more practical for use in the routine clinical laboratory. We used three enzymes in our enzyme kit, 3 #alpha# -hydroxysteroid dehydrogenase (3 #alpha# -HSD), 3-keto-5 #beta# -steroid- #DELTA# ~4-dehydrogenase (#DELTA# ~4-5 #beta# -DH) and diaphorase and three reactions are coupled to enhance the measuring sensitivity.
