A Glucosylated Steroid from the Active Fraction of Indonesian Ficus deltoidea Leaves

Ficus plants are known to contain a variety of alkaloids, terpenoids and flavonoids. We have reported that Indonesian Ficus deltoidea or tabat barito contains methyl 10-epi-pheophorbide A isolated from MCF-7 cells active fraction of the plant. The leaves of F. deltoidea were extracted with aqueous EtOH and was then partitioned into three layers n-hexane, CH_2Cl_2, and n-BuOH. The CH_2Cl_2 layer showed the most toxic layer against zebrafish (Danio rerio) embryos which was agreement with the previous work. Fractionation of the CH_2Cl_2 layer yielded 6 fractions which the most active fraction F caused major malformation of the body axis in zebrafish embryos. Further purification of fraction F with MPLC and HPTLC gave a polar compound 1 named daucosterol, a glucosylated steroid after NMR and HRMS analysis as well as comparing data with literature studies.
