Based on the self-designed and high-throughput microfluidic cell chip and the constructed chip test system, the study tested the effect of the Common Coltsfoot Flower extracts on the process of 5 - lipoxygenase metabolism AA. The designed 3D microfluidic cell chip which consists of 144 microcavities and sandwich cross channels could be directly detected by the commercial Enzyme-linked immunoassay instrument. The fluorescent labeling, washing and detection of suspension cells were integrated in the chip. 5-LOX inhibition ability of five kinds of Common Coltsfoot Flower extracts were tested by the chip. It was illustrated that the extracts of ethyl acetate and petroleum ether layer from Common Coltsfoot Flower could inhibit the active component of 5-LOX, and the activity of candidate materials could be evaluated in vitro effectively. During the process of test, it was also shown that the designed 3D array microfluidic chip demonstrated great advantages in 5-LOX inhibitory activity testing for suspension cell system.
